With Oxford Nanopore’s v14 library preparation chemistry and R10.4.1 flow cells, raw read accuracy typically exceeds 99%. During sequencing, we enforce a minimum raw read Q-score of 10 (approximately 90% accuracy), although most reads achieve Q20 or higher (around 99% accuracy). We also apply a higher-accuracy basecalling model to further enhance data quality.
In the assembly process, reads are filtered to remove short and low-quality sequences. For high-quality DNA samples, the resulting assembled contigs generally reach an accuracy of approximately Q40, equivalent to 99.99%.
Our Hybrid Sequencing service, which incorporates polishing with Illumina data, enables us to achieve even higher accuracy—especially in challenging regions such as error-prone homopolymers and methylated motifs.
